    Development of Walkability Indicator for Smart Shrinking: Case Study of Sprawl Areas in Northern Osaka Metropolitan

    Urban shrinkage is a problem that is faced by some metropolitan areas globally in urban cities faced with depopulation. Therefore, these cities require “smart shrinking”, which consists of design methods intended to satisfy residents’living needs, even though the population is declining. The purpose of this paper is to develop a walkability indicator for visualizing smart shrinking. The methods include statistical analysis by GIS analysis and questionnaire in the Northern Osaka Metropolitan Area where there are many small-sprawl areas. The following four results were identified. First, the population decline in sprawl areas indicates the regional characteristics of each city. Second, this paper develops a walkability indicator composed of household density, convenience of facilities, road connectivity, and traffic safety. Third, Wi(i), which is walkability in each residential area, in sprawl areas, and in public housing areas, is important for future population change. Finally, the validity and effectiveness of the indicator is clarified by questionnaires regarding regional evaluation. In conclusion, the walkability indicator is able to visualize smart shrinking in sprawl areas

    Evaluation of Haplotype Inference Using Definitive Haplotype Data Obtained from Complete Hydatidiform Moles, and Its Significance for the Analyses of Positively Selected Regions

    The haplotype map constructed by the HapMap Project is a valuable resource in the genetic studies of disease genes, population structure, and evolution. In the Project, Caucasian and African haplotypes are fairly accurately inferred, based mainly on the rules of Mendelian inheritance using the genotypes of trios. However, the Asian haplotypes are inferred from the genotypes of unrelated individuals based on population genetics, and are less accurate. Thus, the effects of this inaccuracy on downstream analyses needs to be assessed. We determined true Japanese haplotypes by genotyping 100 complete hydatidiform moles (CHM), each carrying a genome derived from a single sperm, using Affymetrix 500 K Arrays. We then assessed how inferred haplotypes can differ from true haplotypes, by phasing pseudo-individualized true haplotypes using the programs PHASE, fastPHASE, and Beagle. We found that, at various genomic regions, especially the MHC locus, the expansion of extended haplotype homozygosity (EHH), which is a measure of positive selection, is obscured when inferred Asian haplotype data is used to detect the expansion. We then mapped the genome using a new statistic, XDiHH, which directly detects the difference between the true and inferred haplotypes, in the determination of EHH expansion. We also show that the true haplotype data presented here is useful to assess and improve the accuracy of phasing of Asian genotypes

    A novel insertion pathway of mitochondrial outer membrane proteins with multiple transmembrane segments

    The central channel Tom40 of the preprotein translocase of outer membrane (TOM) complex is thought to be responsible for the import of virtually all preproteins synthesized outside the mitochondria. In this study, we analyze the topogenesis of the peripheral benzodiazepine receptor (PBR), which integrates into the mitochondrial outer membrane (MOM) through five hydrophobic transmembrane segments (TMSs) and functions in cholesterol import into the inner membrane. Analyses of in vitro and in vivo import into TOM component–depleted mitochondria reveal that PBR import (1) depends on the import receptor Tom70 but requires neither the Tom20 and Tom22 import receptors nor the import channel Tom40, (2) shares the post-Tom70 pathway with the C-tail–anchored proteins, and (3) requires factors of the mitochondrial intermembrane space. Furthermore, membrane integration of mitofusins and mitochondrial ubiquitin ligase, the MOM proteins with two and four TMSs, respectively, proceeds through the same initial pathway. These findings reveal a previously unidentified pathway of the membrane integration of MOM proteins with multiple TMSs

    本論は,人が生涯にわたり音楽文化とかかわっていく土台を築くため,ブータン歌謡ツァンモの調査から得られた成果を参照しながら,音楽科の新しい方向性を見出すことを目的とした一連の研究に位置づくものである。本年度は,ラジオ放送や学校行事等,現在のツァンモのありように焦点をあて,自分なりの思いを歌で表現する意義を問い直した。とりわけ,歌の含意を共有し,なじみのある詩形や旋律を用いて,掛け合い歌による双方向のコミュニケーションを通して,自分の思いを歌で自在に伝える活動から手がかりを得て,小学校の音楽科授業における実践開発を試みた。To establish a foundation for people to remain involved in musical culture throughout their lives, we sought to find new directions for music lessons at school, referring to the results of a field survey of the Bhutanese tsangmo singing dialogue. In this study, we focused on the present state of tsangmo in radio broadcasting and school events. After referring to Bhutanese singing culture, we applied its dialogue style in music lessons for the lower grades in primary schools. By sharing the meaning and using familiar forms of the song text and melody, children were able to communicate with each other and showed the capacity to develop new learning processes in the playful singing dialogues. This approach may offer us a key to reconsidering the free expression of one's feelings through singing.本研究は,JSPS科研費26301043の助成を受けたものである

    Comparative evaluation of three immunochromatographic identification tests for culture confirmation of Mycobacterium tuberculosis complex

    Background: The rapid identification of acid-fast bacilli recovered from patient specimens as Mycobacterium tuberculosis complex (MTC) is critically important for accurate diagnosis and treatment. A thin-layer immunochromatographic (TLC) assay using anti-MPB64 or anti-MPT64 monoclonal antibodies was developed to discriminate between MTC and non-tuberculosis mycobacteria (NTM). Capilia TB-Neo, which is the improved version of Capilia TB, is recently developed and needs to be evaluated.Methods: Capilia TB-Neo was evaluated by using reference strains including 96 Mycobacterium species (4 MTC and 92 NTM) and 3 other bacterial genera, and clinical isolates (500 MTC and 90 NTM isolates). M. tuberculosis isolates tested negative by Capilia TB-Neo were sequenced for mpt64 gene.Results: Capilia TB-Neo showed 100% agreement to a subset of reference strains. Non-specific reaction to M. marinum was not observed. The sensitivity and specificity of Capilia TB-Neo to the clinical isolates were 99.4% (99.6% for M. tuberculosis, excluding M. bovis BCG) for clinical MTC isolates and 100% for NTM isolates tested, respectively. Two M. tuberculosis isolates tested negative by Capilia TB-Neo: one harbored a 63-bp deletion in the mpt64 gene and the other possessed a 3,659-bp deletion from Rv1977 to Rv1981c, a region including the entire mpt64 gene.Conclusions: Capilia TB-Neo is a simple, rapid and highly sensitive test for identifying MTC, and showed better specificity than Capilia TB. However, Capilia TB-Neo still showed false-negative results with mpt64 mutations. The limitation should be recognized for clinical use
